EnSys is the pioneer in the development of rapid analytical capabilities. EnSys analytical methods can be used instead of traditional testing methods to rapidly map the extent of contamination or monitor the removal of contaminated matrix during evaluation. This technology delivers rapid, accurate indications of contamination in soil, water, surfaces and waste organic liquids. EnSys tests kits are an on-demand system, and completely portable. The EnSys system includes tests methods that are developed and rigorously tested to ensure a high level of accuracy in the field or laboratory.
Immunoassay technology relies on an antibody that is developed to have a high degree of sensitivity to the target compound. This antibody's high specificity is coupled with a sensitive colorimetric reaction that provides a visual result.
THE ENSYS IMMUNOASSAY PROCESS:
1. The antibody, specific for the target analyte, is coated on a test tube.
2. The target analyte, which may be present in the samples, "competes" with the enzyme conjugate for a limited number of antibody binding sites.
3. A washing step eliminates any target analyte or enzyme conjugate that has not bound to the antibody in the tube.
4. The substrate solutions are added and react in the presence of the enzyme conjugate to produce color.
5. The intensity of the color formed is inversely proportional to the concentration of the target analyte in the sample.
6. The absence/presence determination is made by comparing the color developed by a sample of unknown concentration to the color formed with the standard containing the analyte at a known concentration.
EnSys claims the tests correctly identify 95% of the samples that contain concentrations at or above the detection level. EnSys further claims the tests correctly identify 95% of the samples that contain no contamination. Through the development of environmental applications for accurate, reliable test technologies EnSys rapid test methods have been accepted by the Environmental Protection Agency (EPA), under the Office of Solid Waste methods development program. EnSys currently has seven rapid tests accepted under EPA methods:
Pentachlorophenol (soil, water), EPA Method 4010
PCB (soil, organic liquids), EPA Method 4020
Petroleum fuels (soil), EPA Method 4030
Polyaromatic hydrocarbons (soil), EPA Method 4035
Nitroaromatics (soil), EPA Method 8515
Limitations:
The immunoassay technology is generally specific in its detection capabilities, thereby prohibiting detection of broad groups of compounds, i.e. the immunoassay detects polychlorinated biphenyls but not all chlorinated compounds. However, the antibody might recognize a compound chemically very similar to the target compound causing false positives results. Immunoassay technology is not applicable for all classes of small, environmentally significant molecules; for example, long chain aliphatic hydrocarbon compounds and most transition metals. Temperature extremes can affect test results; the operational temperature ranges are typically between 40F and 100F. The analysis can be affected by specific matrix conditions. For example, analyte recovery for analytes like PCB and PAH can be affected by excessive soil moisture. Additionally, percent levels of oil can result in false positive results.
